(EP2500420) The present invention relates to a polynucleotide encoding an omega 3 (w-3) desaturase from Pythium irregulare with specificity to long chain polyunsaturated omega 6 (É-6) fatty acids as well as a vector containing said polynucleotide, and a host cell containing the vector or the polynucleotide.  Moreover, the present invention pertains to a polypeptide encoded by the said polynucleotide, antibodies against the polypeptide as well as a method for the manufacture of the polypeptide.  Further, encompassed by the present invention are transgenic non-human organisms.  Finally, the present invention relates to methods for the manufacture of compounds and oil- fatty acid- or lipid-containing compositions.

(EP2500420) 1. A cell capable of producing w-6 docosapentaenoic acid (DPA), further comprising a heterologous omega -3 desaturase capable of converting w-6 DPA into docosahexaenoic acid (DHA). 2. A non-human organism, preferably an animal, plant or plant part or a multicellular microorganism, comprising or consisting of the cell of claim 1. 3. A plant part comprising a heterologous omega -3 desaturase polypeptide, wherein the w-3 desaturase polypeptide a) has an amino acid sequence of at least 75% identity to the amino acid sequences shown in SEQ ID NO: 2 or 24, or b) has an amino acid sequence coded (a) by a nucleic acid being a fragment of a nucleic acid sequence as shown in SEQ ID NO. 1 or 23, or (b) by a nucleic acid which is at least 75% identical to a nucleic acid sequence coding for to the amino acid sequences shown in SEQ ID NO: 2 or 24, orc) is capable of converting w-6 docosapentaenoic acid (DPA) into docosahexaenoic acid (DHA), wherein said polypeptide comprises a polypeptide pattern as shown in a sequence selected from the group consisting of SEQ ID NO: 15, 16, 17, 18, 19, 20, 21, 22, 37, 38, 39, 40, 41, 42, 43, 44 and 45, or d) is obtainable by expression of a nucleic acid coding for a polypeptide according to any of a) to c) in a plant cell. 4. The plant part according to claim 2 or 3, wherein the plant part is selected from leaf, stem, seed, root, tubers, anthers, fibers, root hairs, stalks, embryos, calli, cotelydons, petioles, harvested material and plant tissue, preferably a seed. 5. The plant part according to any of claims 2 to 4, wherein the plant part comprises a cell which additionally comprises at least one further enzyme being involved in the biosynthesis of fatty acids or lipids, and wherein said further enzyme is preferably selected from the group consisting of acyl-CoA dehydrogenase(s), acyl-ACP [= acyl carrier protein] desaturase(s), acyl-ACP thioesterase(s), fatty acid acyltransferase(s), acyl-CoA:lysophospholipid acyltransferase(s), fatty acid synthase(s), fatty acid hydroxylase(s), acetyl-coenzyme A carboxylase(s), acyl-coenzyme A oxidase(s), fatty acid desaturase(s), fatty acid acetylenase(s), lipoxygenase(s), triacylglycerol lipase(s), allenoxide synthase(s), hydroperoxide lyase(s) or fatty acid elongase(s), acyl-CoA:lysophospholipid acyltransferase, DELTA 4-desaturase, DELTA 5-desaturase, DELTA 6-desaturase, DELTA 8-desaturase, DELTA 9-desaturase, DELTA 12-desaturase, DELTA 5-elongase, DELTA 6-elongase and DELTA 9-elongase. 6. A polynucleotide comprising a nucleic acid sequence encoding a polypeptide having omega -3 desaturase activity and being capable of converting w-6 docosapentaenoic acid (DPA) into docosahexaenoic acid (DHA), wherein said polypeptide comprises a polypeptide pattern as shown in a sequence selected from the group consisting of SEQ ID NO: 15, 16, 17, 18, 19, 20, 21, 22, 37, 38, 39, 40, 41, 42, 43, 44 and 45. 7. A vector comprising the polynucleotide of claim 6, wherein the vector preferably is an expression vector. 8. The vector of claim 7, wherein said vector comprises at least one polynucleotide encoding a further enzyme being involved in the biosynthesis of fatty acids or lipids, and wherein said further enzyme is preferably selected from the group consisting of acyl-CoA dehydrogenase(s), acyl-ACP [= acyl carrier protein] desaturase(s), acyl-ACP thioesterase(s), fatty acid acyltransferase(s), acyl-CoA:lysophospholipid acyltransferase(s), fatty acid synthase(s), fatty acid hydroxylase(s), acetyl-coenzyme A carboxylase(s), acyl-coenzyme A oxidase(s), fatty acid desaturase(s), fatty acid acetylenase(s), lipoxygenase(s), triacylglycerol lipase(s), allenoxide synthase(s), hydroperoxide lyase(s) or fatty acid elongase(s), acyl-CoA:lysophospholipid acyltransferase, DELTA 4-desaturase, DELTA 5-desaturase, DELTA 6-desaturase, DELTA 8-desaturase, DELTA 9-desaturase, DELTA 12-desaturase, DELTA 5-elongase, DELTA 6-elongase and DELTA 9-elongase. 9. A method for the manufacture of a polypeptide having w-3 desaturase activity comprising: a) expressing the polynucleotide of claim 6 in a host cell; and b) obtaining the polypeptide encoded by said polynucleotide from the host cell. 10. A polypeptide encoded by the polynucleotide of claim 6 or which is obtainable by the method of claim 9. 11. An antibody which specifically recognizes the polypeptide of claim 10. 12. A method for the manufacture of a compound having a structure as shown in the general formula I (see diagramm) wherein the variables and substituents in formula I are R **1 = hydroxyl, coenzyme A (thioester), lysophosphatidylcholine, lysophosphatidylethanolamine, lysophosphatidylglycerol, lysodiphosphatidylglycerol, lysophosphatidylserine, lysophosphatidylinositol, sphingo base or a radical of the formula II (see diagramm)R **2 = hydrogen, lysophosphatidylcholine, lysophosphatidylethanolamine, lysophosphatidylglycerol, lysodiphosphatidylglycerol, lysophosphatidylserine, lysophosphatidylinositol or saturated or unsaturated C2-C24-alkylcarbonyl, R **3 = hydrogen, saturated or unsaturated C2-C24-alkylcarbonyl, or R2 and R3 independently of each other are a radical of the formula la:(see diagramm) n = 2, 3, 4, 5, 6, 7 or 9, m = 2, 3, 4, 5 or 6 and p = 0 or 3; and  wherein said method comprises cultivating the cell, non-human organism or plant part according to any of claims 1 to 5 under conditions which allow biosynthesis of the said compound. 13. A method for the manufacture of an oil-, fatty acid- or lipid-containing composition comprising the steps of the method of claim 12 and the further step or formulating the compound as an oil-, fatty acid- or lipid-containing composition. 14. The method of claim 13, wherein said oil-, fatty acid or lipid-containing composition is further formulated as a pharmaceutical composition, a cosmetic composition, a foodstuff, a feedstuff, a fish feed or a dietary supply. 15. Use of the polynucleotide of claim 6, the vector of any one of claims 7 or 8, the cell of claim 1, the polypeptide of claim 10 or the non-human organism or plant part of any of claims 2 to 5 for the manufacture of an oil-, fatty acid- or lipid-containing composition. 16. The use of claim 15, wherein said oil-, fatty acid- or lipid-containing composition is to be used as a pharmaceutical composition, cosmetic composition, foodstuff, feedstuff, fish feed or dietary supply. 17. Use of the polynucleotide of claim 6, the vector of any one of claims 7 or 8, the cell of claim 1, the polypeptide of claim 10 or the non-human organism or plant part of any of claims 2 to 5 for converting w-6 DPA into DHA or increasing the content of DHA in a cell or in a plant part, preferably in a plant seed. 18. Borage oil, calendula oil, canola oil, castor-oil oil, coconut oil, cotton oil, evening primrose oil, hemp oil, linseed oil, mullein oil, mustard oil, oil palm oil, oilseed rape oil, olive oil, peanut oil, poppy oil, pumpkin/squash oil, Punica oil, safflower oil, sesame oil, soybean oil, sunflower oil, thistle oil, tobacco oil, walnut oil, obtainable by extracting a respective plant part according to any of claims 2 to 5. 19. The canola oil according to claim 18 obtainable by extracting a seed of a canola plant according to any of claims 2 to 5.
