(IN201621023343) “CHIMERIC DELTA ENDOTOXIN PROTEIN WITH IMPROVED INSECTICIDAL ACTIVITY” The present invention relates to develop a chimeric delta endotoxin protein, a novel hybrid Bt toxin constructed by domain shuffling between Cry1Ac and Cry9Aa.  Herein, the novel Bt hybrid construct of chimeric delta endotoxin protein is having coding nucleic acid sequence of SEQ ID No. 1, encoding the said chimeric protein, which is having an extra ordinary insecticidal property with unique domain election from the parent Cry toxins i.e.  Cry1Ac and Cry9Aa.  And to provide potent insecticidal hybrid Bt toxin turning out to be an excellent biocontrol agent.

An insecticidal chimeric delta endotoxin Cry1Ac-Cry9Aa having coding nucleic acid sequence of SEQ ID NO: 1 constructed by domain shuffling between Cry9Aa and Cry1Ac, wherein Cry1Ac-Cry9Aa comprises Domain I of Cry1Ac and Domain II and Domain III of Cry9Aa. 2. The insecticidal chimeric delta endotoxin Cry1Ac-Cry9Aa as claimed in claim 1, wherein said domains of respective Cry toxins are amplified by PCR amplification. 3. The insecticidal chimeric delta endotoxin Cry1Ac-Cry9Aa as claimed in claim 1, wherein said Cry1Ac-Cry9Aa is constructed by overlap extension PCR mediation recombination, herein a short overlapping sequence of six base pairs is designed with one mismatch in homologous sequence to recombine the domain coding regions of Cry1Ac and Cry9Aa genes. 4. The insecticidal chimeric delta endotoxin Cry1Ac-Cry9Aa as claimed in claim 1, wherein said chimeric construct gave 4.9 fold enhanced toxicity to H. armigera when tested with Cry1AC as control. 5. The insecticidal chimeric delta endotoxin Cry1Ac-Cry9Aa as claimed in claim 1, wherein said chimera has a length of 1893 base pairs (bp). 6. The insecticidal chimeric delta endotoxin Cry1Ac-Cry9Aa as claimed in claim 1, wherein said chimeric nucleic acid has ATGGAT at its 5' extreme, as an initiation codon. 7. The insecticidal chimeric delta endotoxin Cry1Ac-Cry9Aa as claimed in claim 1, wherein said chimeric nucleic acid has AGTAG as a translation termination codon. 8. The insecticidal chimeric delta endotoxin Cry1Ac-Cry9Aa as claimed in claim 1, wherein said chimeric nucleic acid construct is constructed with restriction enzymes NheI and XhoI, added at flanking ends of gene.
